RHD gene deletion occurred in the Rhesus box.

چکیده
The Rh blood group antigens derive from 2 genes, RHD and RHCE, that are located at chromosomal position 1p34.1-1p36 (chromosome 1, short arm, region 3, band 4, subband 1, through band 6). In whites, a cde haplotype with a deletion of the whole RHD gene occurs with a frequency of approximately 40%. The relative position of the 2 RH genes and the location of the RHD deletion was previously unknown. A model has been developed for the RH locus using RHD- and RHCE-related nucleotide sequences deposited in nucleotide sequence databases along with polymerase chain reaction (PCR) and nucleotide sequencing. The open reading frames of both RH genes had opposite orientations. The 3' ends of the genes faced each other and were separated by about 30 000 base pair (bp) that contained the SMP1 gene. The RHD gene was flanked by 2 DNA segments, dubbed Rhesus boxes, with a length of approximately 9000 bp, 98.6% homology, and identical orientation. The Rhesus box contained the RHD deletion occurring within a stretch of 1463 bp of identity. PCR with sequence-specific priming (PCR-SSP) and PCR with restriction fragment length polymorphism (PCR-RFLP) were used for specific detection of the RHD deletion. The molecular structure of the RH gene locus explains the mechanisms for generating RHD/RHCE hybrid alleles and the RHD deletion. Specific detection of the RHD(-) genotype is now possible. Investigation of Paternal RhD Zygosity by Two Molecular Methods among Blood Donors in Kurdistan Province, Iran
Background: RhD antigen system is the leading cause of hemolytic disease of the fetus and newborn (HDFN). Paternal molecular RhD zygosity test is valuable to decide on the use of anti-D immunoglobulin prophylaxis in Rh D-negative pregnant women. We aimed to investigate the paternal RhD zygosity by two molecular methods among blood donors in Kurdistan province, the west of Iran. We also compared...
متن کاملDetermination of RhD zygosity: comparison of a double amplification refractory mutation system approach and a multiplex real-time quantitative PCR approach.
BACKGROUND Rh isoimmunization and hemolytic disease of the newborn still occur despite the availability of Rh immunoglobulin. For the prenatal investigation of sensitized RhD-negative pregnant women, determination of the zygosity of the RhD-positive father has important implications. The currently available molecular methods for RhD zygosity assessment, in general, are technically demanding and...
متن کاملجداسازی و شناسایی آنتیژن RhD از غشای گلبول قرمز به روش رسوب ایمنی
Background and Objective: Rh (Rhesus) is a highly complex blood group system in man which plays an important role in transfusion medicine. The aim of this study was the isolation of RhD protein from the membrane of RBCs. Materials and Methods: In this experimental study immunoprecipitation method with human anti-RhD polyclonal antibody was utilized for the isolation of RhD antigen from Rh+ huma...
متن کاملIdentification of a partial internal deletion in the RH locus causing the human erythrocyte D--phenotype.
The D--phenotype of the human erythrocyte is a genetic variant of the Rh blood group system associated with the expression of D but not C, c, E, and e (designated non-D) antigens. In this report, we characterize the structure and expression of Rh polypeptide genes in two D--homozygotes of Italian origin. Southern blot analysis detected a gross deletion in their genomic DNA that correlated with ...
متن کاملThe fetal and neonatal outcomes of Rhesus D antibody affected pregnancies in Northern Ireland.
It has been suggested that routine antenatal prophylactic anti-D should be introduced for prevention of Rhesus D (RhD) haemolytic disease. Before making changes to the current prevention program it is important, therefore, to have up-to-date data on affected infants. Pregnant women with anti-D antibodies between September 1994 and February 1997 were identified by the Northern Ireland Blood Tran...
